She is best known for her many film roles, such as Vickie Miner in Reality Bites, Abby Barnes in The Truth About Cats and Dogs, Marcy Tizard in The Matchmaker, Anita Hoffman in Steal This Movie!, and Hannah Rosenbloom in Love Hurts. She also starred in various TV shows, such as NewsRadio, Ellen, Seinfeld, Home Improvement, Law & Order, The West Wing, and Criminal Minds: Suspect Behavior. Additionally, she voiced Stith in Don Bluth's sci-fi animated film Titan A.E. and played Minnie Mogul in the Universal Pictures hybrid film The Adventures of Rocky and Bullwinkle (both released in 2000).

For Disney, she voiced Ursula in the English dub of Studio Ghibli's Kiki's Delivery Service, Bridget in the 2006 animated feature film The Wild, and Colette Tatou in the 2007 Disney/Pixar film Ratatouille. She also played Marissa in Flora & Ulysses, Heather Mooney in the 1997 Touchstone Pictures film Romy and Michele's High School Reunion, Officer Monica Romero in Big Trouble, and Janet Withers in the ABC TV show Stumptown.

Garofalo was born in Newton, New Jersey. While studying history at Providence College, Garofalo entered a comedy talent search sponsored by the Showtime cable network, winning the title of "Funniest Person in Rhode Island". Her original gimmick was to read off her hand, which was not successful in subsequent performances. Dreaming of earning a slot on the writing staff of the TV show Late Night with David Letterman, she became a professional stand-up comedian upon graduating with degrees in history and American studies.
